Detailed data on research infrastructure/laboratory

  1.  Description and data on the research infrastructure/laboratory:
The laboratory represents a comprehensive complex that is sufficient for common applications of molecular biology and genetics for research focused on solving problems in the field of selection of reproductive material in times of climate change, various genetically conditioned physiological responses to changes in natural conditions and stress factors, various population-genetic aspects of management of some endangered or protected species of plants and animals. Since the methodologies and laboratory equipment that we currently use and have been no longer sufficient for the study of some aspects, we also provide solutions for the use of methodologies that provide a higher number of genetic markers, which expand the portfolio of problems that can be solved in a forestry-oriented laboratory. In the future, we would like to prepare libraries for NGS (next generation sequencing) ourselves, but we would still need investments in additional devices for assessing the quantity and quality of nucleic acids and for the selection of fragments for sequencing, which we currently also provide solutions for. We also needed additional computing capacity for calculations that currently take more than a few days, sometimes even weeks.
  1. List of equipment:

  • Autoclaves
  • Nanodrop spectrophotometer
  • Qubit fluorimeter
  • Lyophilizer
  • ABI 3130 capillary genetic analyzer
  • PCR thermocyclers: Biometra T Personal, Eppendorf, real-time PCR Biorad iQ5
  • Eppendorf heat sealer
  • Eppendorf deep-freezing box
  • laminar flow boxes
  • ice maker
  • Eppendorf Comfort 5355 thermomixer 3 pcs
  • Retsch MM 301 oscillating mill
  • various laboratory centrifuges
  • various apparatus for vertical and horizontal electrophoresis
  • common laboratory equipment pH meters, vortexes, scales, pipettes, shakers, demineralized and ultrapure water generators
  1. Justification of uniqueness/strategic importance:
The Population Genetics Laboratory is a unique laboratory dedicated to the population genetics of forest trees and wildlife with a focus on forestry and conservation applications. Among the forestry topics, the topic of forest reproductive material dominates, also in relation to climate change, provenance research in various contexts, the impact of forestry measures on the genetic diversity of forest trees and forest communities. In conservation topics, we mainly focused on the genetic diversity and differentiation of wildlife - bear, wolf, deer, capercaillie, marmot, ground squirrel and the application of genetic aspects in the protection of the above-mentioned species. In the laboratory, we also deal with some classic biological topics - selection, hybridization, phylogenesis, phylogeography, etc., especially in relation to forest trees and wildlife. Our department participated in the draft law on forest reproductive material, in the development of management plans for protected animal species - capercaillie, bear, otter, chamois, marmot.
